Abstract
A simple spectrophotometric assay for diamine oxidase (histaminase) has been modified
for the measurement of this enzyme in sera. It depends upon the enzymatic conversion
of diaminobutane to Δ1-pyrroline and the condensation of this product with o-amino-benzaldehyde to yield
a yellow chromophore. Serum levels of diamine oxidase have been determined during
normal pregnancy in forty-five women. This enzymatic activity increases sharply early
in the second trimester of pregnancy and is maximal early in the third trimester.
No diamine oxidase activity is detectable in sera from nonpregnant individuals. On
the other hand, serum levels of monoamine oxidase, as measured by the enzymatic oxidation
of benzylamine, do not change during normal pregnancy and are not different from levels
obtained in nonpregnant individuals. These data are presented to indicate that sera
from pregnant women contain two independent amine oxidases.
