The preparation of polycythemic mice by the use of silicone rubber membranes for erythropoietin bioassays is reported. The results of hematocrit values, reticulocyte counts, red blood cell 59Fe incorporations, and responses to erythropoietin illustrate that these animals are quite similar in their erythropoietin responses to animals made polycythemic by other techniques. The results presented in this report indicate that for optimum erythropoietin bioassays, mice should be housed, 11 mice per cage, for 2 to 3 weeks in membrane cages and assayed 6 to 12 days after their removal. Of the mouse strains investigated, the C3H/HeJ mouse appears to be the most sensitive to erythropoietin after removal from the membrane cages.
